چکیده

We propose a simple model for the kinetics of packaging of viral DNA into a capsid against an external force trying to prevent it. The model leads to a Butler-Volmer-type dependence of the rate of packaging on the pulling force F.

Generation of Helper Plasmids Encoding Mutant Adeno-associated Virus Type 2 Capsid Proteins with Increased Resistance against Proteasomal Degradation

Objective(s): Adeno-associated virus type 2 (AAV2) vectors are widely used for both experimental and clinical gene therapy. A recent research has shown that the performance of these vectors can be greatly improved by substitution of specific surface-exposed tyrosine residues with phenylalanines. In this study, a fast and simple method is presented to generate AAV2 vector helper plasmids encod...

Viral DNA Packaging: One Step at a Time

During its life-cycle the bacteriophage ϕ29 actively packages its dsDNA genome into a proteinacious capsid, compressing its genome to near crystalline densities against large electrostatic, elastic, and entropic forces. This remarkable process is accomplished by a nano-scale, molecular DNA pump – a complex assembly of three protein and nucleic acid rings which utilizes the free energy released ...

A Promiscuous DNA Packaging Machine from Bacteriophage T4

Complex viruses are assembled from simple protein subunits by sequential and irreversible assembly. During genome packaging in bacteriophages, a powerful molecular motor assembles at the special portal vertex of an empty prohead to initiate packaging. The capsid expands after about 10%-25% of the genome is packaged. When the head is full, the motor cuts the concatemeric DNA and dissociates from...

Investigation of viral DNA packaging using molecular mechanics models.

A simple molecular mechanics model has been used to investigate optimal spool-like packing conformations of double-stranded DNA molecules in viral capsids with icosahedral symmetry. The model represents an elastic segmented chain by using one pseudoatom for each ten basepairs (roughly one turn of the DNA double helix). Force constants for the various terms in the energy function were chosen to ...

Stuffing a virus with DNA: dissecting viral genome packaging.

I t is rare that a fully functional biological system presents a simple biophysical problem. The problem of viral DNA packaging comes tantalizingly close: What forces are required to stuff double-stranded DNA of fixed length into a container of fixed volume? Now that biophysicists have developed the capability to measure such forces, theoretical solutions to this problem can be directly tested,...
